"I exposed my ethidium bromide-stained gel to UV light, only to discover a galaxy of speckles (still not sure why...) and no PCR products. :(. But on the bright side, I think I can see Orion's belt!"

You don't know how to make Jello?
ReplyDeleteIs this a DNA or RNA gel? This is too compelling a mystery to ignore.
ReplyDeleteDNA gel.
ReplyDeletedid you run the gel with pcr products in the wells, or did you mix the pcr products in the agar before it set? :)
ReplyDeleteHow old is the buffer you stored your gel in? I'm betting your EtBr is lighting up bacterial contamination.
ReplyDeleteThat usually means you didn't let the agar melt properly before you poured it out to set. The speckles are the bits of unmelted agar powder. Why, yes, I have done that myself a few times. =)
ReplyDeleteLooks like someone has reused their agar a few too many times (^_^);;
